| dc.description.abstract | Digital transformation in healthcare increasingly integrates artificial intelligence (AI), and computational modelling, to complement traditional experimental validation of medical devices. Total knee replacement (TKR) implants, require extensive mechanical, and tribological evaluation, which remains time-intensive and resource-demanding. This study developed and validated a quantitative, physics-informed, data-driven digital framework for generating synthetic biomechanical data and predicting stress, deformation, and wear behaviour in modular all-polymer knee trays manufactured from PEEK-OPTIMA. A synthetic dataset, comprising 8000 design configurations, was generated using Latin Hypercube Sampling, across literature-supported geometric, material, and loading ranges. Stress and deformation, were estimated using simplified contact mechanics relationships, while wear was modelled using Archard's wear law. Supervised regression models, including Random Forest, Gradient Boosting, and Multilayer Perceptron networks, were trained to predict biomechanical responses. Ensemble tree-based models achieved high predictive accuracy for stress and deformation. Wear prediction demonstrated moderate explanatory power, reflecting the nonlinear and cumulative nature of tribological processes. Validation against ISO 14243 loading standards and multi-design OrthoLoad in vivo envelopes confirmed physiological plausibility. The framework demonstrated that physics-informed synthetic data, combined with machine learning, can support early-stage digital screening of implant designs. | |
